Comparative Study of the Effects of Methyl Jasmonate and Abscisic Acid on Rna and Protein Synthesis in Excised Cotyledons of Cucurbita

چکیده
The effects of methyl jasmonate and abscisic acid on the polypeptide pattern of soluble proteins as well as in vivo protein and RNA synthesis and endogenous nuclear RNA polymerase activity were compared in excised marrow cotyledons grown in the light. Treatment of cotyledons with MeJA resulted in the induction of two abundant polypeptide bands with Mr 43 and 60 kDa and a significant accumulation of a polypeptide band with Mr 97.4 kDa as analysed by SDS–PAGE. In contrast to MeJA, ABA did not cause the formation of these specific polypeptide bands. Furthermore, the inhibitory effect of ABA on protein and RNA biosynthesis as well as the activity of total endogenous RNA polymerases was much stronger than the effect of MeJA. It is suggested that in excised marrow cotyledons MeJA and ABA act by different mechanisms in the regulation of cotyledon growth and development during the earlier stages of germination.

INTERACTION BETWEEN METHYL ESTER OF JASMONIC ACID AND BENZYLADENINE DURING THE GROWTH OF EXCISED GREENING COTYLEDONS OF Cucurbita pepo L. (zucchini)
Treatment of excised marrow cotyledons (Cucurbita pepo L. (zucchini) growing under photoperiod with methyl ester of jasmonic acid (MeJA) resulted in a decrease of their fresh weight. This effect did not depend on the exogenously applied MeJA concentrations. Furthermore, in vivo labelling experiments showed an inhibition of the total protein synthesis. On the other hand, specific quantitative ch...
متن کاملEffect of Methyl Ester of Jasmonic Acid, Abscisic Acid and Benzyladenine on Chlorophyll Synthesis in Excised Cotyledons of Cucurbita
Except for the first stages of illumination, exogenous methyl ester of jasmonic acid (MeJA) inhibited much stronger than abscisic acid (ABA) the accumulation of chlorophyll in excised cotyledons of C. pepo L. (zucchini) exposed to controlled light conditions. Benzyladenine (BA) applied in a combination with MeJA or ABA at equimolar concentrations completely neutralized their inhibitory effect. ...

The jasmonates are well studied in the context of plant defence but increasingly are also recognised as playing roles in development. In many systems, jasmonates antagonise the effects of cytokinins. The aim of the present work was to elucidate interactions between methyl jasmonate and cytokinin (benzyladenine) in regulating growth of zucchini (Cucurbita pepo L., cv. Cocozelle, var. Tripolis) c...
متن کاملThe effect of methyl jasmonate on antioxidant enzymes and improving the resistance of Cucurbita pepo in low temperature
Early sowing in spring season is important for the use of mild conditions as well as proper vegetative growth, but the occurrence of low temperatures at the beginning of the growing season may cause the problem for plant establishment. In order to investigate the effect of methyl jasmonate (MeJA) on antioxidant enzymes and improving the resistance of pumpkin (Cucurbita pepo) to low temperature ...
متن کاملHormonal Regulation of Ribosomal Rna Gene Expression in Excised Cotyledons of Cucurbita
In the present work we have studied the effects of exogenously applied methyl ester of jasmonic acid (MeJA) and benzyladenine (BA) on rRNA gene expression in the model system of excised marrow cotyledons (Cucurbita pepo L. zucchini). Treatment of cotyledons with BA or MeJA resulted in a 2-fold increase and a 25–30% decrease in the endogenous nuclear RNA polymerase I activity, respectively. The ...
